【摘要】 構(gòu)建了香菇135菌株(Lentinula edodes)菌絲生物量與其DNA含量的標準曲線。通過一系列的篩選工作,確定了標準菌絲的培養(yǎng)條件及DNA提取系統(tǒng),得到了三個不同培養(yǎng)批次香菇135菌絲體構(gòu)建的高重現(xiàn)性的關聯(lián)標準曲線,它們均具有較高的相關系數(shù)(>0.99),統(tǒng)計構(gòu)建標準曲線的所有90個數(shù)據(jù),測定的單點誤差范圍為0.05%13.74%(<14%)。所得數(shù)據(jù)表明在操作范圍內(nèi),菌絲生物量與DNA含量之間存在很好的線性關系,每mg香菇135標準培養(yǎng)菌絲(干重)對應的DNA含量為6.1μg。
【Abstract】 Standard curves relating DNA content and mycelial biomass were constructed for Lentinula edodes,strain 135.Adopting a set of standardized mycelium growth conditions and DNA extraction procedures,standard curves relating DNA concentration with mycelial biomass for three separate batches of L.edodes,strain 135,exibited a correlation coefficient >0.99.The error range for a total of 90 determinations(18 samples,five replicates of each sample)was between 0.05%-13.74%.Our data indicated that,under ...
