【中文摘要】 以苯酚硫酸法作為靈芝多糖的測定方法,研究了從固體發(fā)酵靈芝菌絲體中提取水溶性多糖的工藝,對浸提過程中的溫度、時間、固液比及提取次數(shù)4個因素進行了單因素試驗和正交試驗,并利用SephadexG-100凝膠色譜法和薄層層析法對主要組分GLPⅠ進行了分子量測定和單糖分析。結(jié)果表明,靈芝多糖的最佳提取條件為浸提溫度95℃,浸提時間40min,固液比1∶20和提取次數(shù)為2次;GLPⅠ分子量約為179900,初步判斷為甘露聚糖。
【英文摘要】 Using phenol-sulphuric acid method to detect the contents of polysaccharides, the extraction of the water-soluble polysaccharides from Ganoderma lucidum mycelia was studied. The optimum extraction conditions were determined by single-factor test and orthogonal test. And the molecular weight and monosaccharide composition of GLPⅠ were determined by Sephadex G-100 gel filtration chromatography and thin-layer chromatography. The results showed that the optimum extracting temperature, time, ratio of mycelia to water ( w∶v ) and extracting times were 95℃, 40 min, 1∶20 and 2 times. The molecular weight of GLPⅠ was approximately 179 900. GLP I was probably composed of D-mannose.
