反相高效液相色譜法測定瓦尼木層孔菌中柚皮素的含量

【摘要】 采用反相高效液相色譜法(RP-HPLC)測定瓦尼木層孔菌(Phellinus vaninii)中柚皮素含量。瓦尼木層孔菌醇提物水解后采用RP-HPLC對柚皮素的含量進行測定,色譜條件:Hypersil ODS(4.6mm×100mm,3μm)色譜柱,流動相為1%冰醋酸溶液-甲醇(62∶38),柱溫為40℃,流速為0.6mL/min,檢測波長為288nm。標準曲線為A=4.18×106 X+40109(r=0.9998,n=5),柚皮素進樣量在0.0202～2.020μg間線性關系良好,相對標準偏差(RSD)為1.56%(n=5),回收率為97.8%,重現(xiàn)性實驗RSD為1.76%。該法適于瓦尼木層孔菌醇提物中柚皮素的含量測定。

【Abstract】 A method for determining naringenin levels in Phellinus vaninii fruit bodies by reversed-phase highperformance liquid chromatography(RP-HPLC)is described.Ethanolic(70v/v)extracts of P.vaninii fruit bodies were treated with 2% H2SO4at 100℃for 30mins.After adjusting the pH to 7.0with 40% NaOH,the solution was extracted with ethyl acetate and the extracts were freeze-dried.Samples(400 mg)of the freeze-dried material were dissolved in 25mL methanol,aliquots(20μL)of the solution were applied to a Hyp...
